研究领域

  造血干细胞在基因治疗领域具有巨大的应用潜力,因为基因改良后的造血干细胞不仅能够自我更新,而且能够分化成各种血细胞和免疫细胞,可替换原有造血和免疫系统。临床上,造血干细胞移植不仅广泛应用于血液系统疾病以及自身免疫疾。谄渌堤辶龅闹瘟浦,比如生殖细胞瘤,乳腺癌,小细胞肺癌上也被应用于常规治疗失败或复发难治以及具有不良预后因素的患者。基因编辑领域,特别是CRISPR的快速发展,为有效治疗单基因遗传病指明了方向,使单基因遗传病治愈变得可能。对于大部分的疾病来说,靶向性的基因修复是治疗疾病的唯一方案。然而在慢速分裂的造血干细胞中,效率是非常低的。提高造血干细胞中的同源重组效率,达到精准修复基因突变,是开发针对性的基因编辑疗法的关键。本课题组综合应用分子生物学、生物信息学、病毒学,模式动物,细胞学和遗传学的手段来解决此难点,主要包括:

  1. 造血干细胞中同源重组的分子调控

  2. 利用CRISPR为基础的编码系统,建立单细胞标记示踪造血干细胞的新方法

  3. 溶酶体储积症的新型基因治疗
